NLRP3 inflammasome as a novel target for cystic fibrosis treatment

چکیده

Abstract Background Inflammasomes are intracellular multiprotein complexes that sense danger signals from damaged cells and pathogens assemble to mediate caspase-1 activation, which results in the proteolytic cleavage of pro-IL-1β IL-18 into bioactive forms. The NOD-like receptor family pyrin domain containing 3 (NLRP3) inflammasome is a critical component innate immune system mediates activation secretion proinflammatory cytokines IL-1β/IL-18 response disturbances cellular homeostasis caused by microbial infections damage. Main body abstract NLRP3 associated with various inflammatory disorders, including Alzheimer’s disease, diabetes, atherosclerosis. In recent years, has also been implicated inflammation cystic fibrosis. differentiation pro-IL-1β–IL-1β, an active cytokine, mediated neutrophil expression inflammasome. Furthermore, it maintains cytokine storm lungs during pathogenesis CF. Short conclusion This review highlights metabolic reprogramming characterized Warburg effect, NLRP3-mediated fibrosis, its inhibition as potential therapeutic strategy.

NLRP3 inflammasome: a new target in major depressive disorder.

Inflammasome has emerged recently as an unexpected sensor for metabolic danger and stress. Indeed, it has been implicated in the development of major diseases such as gout, type 2 diabetes, and obesity-induced insulin resistance and is increasingly suspected of playing a major role in other human pathologies such as cancer, asbestosis, and Alzheimer’s disease. The inflammasome is a protein comp...

Inflammasome as a Therapeutic Target for Cancer Prevention and Treatment

Chronic inflammation is a critical modulator of carcinogenesis through secretion of inflammatory cytokines, which leads to the formation of an inflammatory microenvironment. In this process, the inflammasome plays an important role in the expression and activation of interleukin (IL)-1β and IL-18 to promote cancer development. The inflammasome is a multiprotein complex consisting of several nuc...

Innate immunity mediated by TLR5 as a novel antiinflammatory target for cystic fibrosis lung disease.

Novel therapies to target lung inflammation are predicted to improve the lives of people with cystic fibrosis (CF) but specific antiinflammatory targets have not been identified. The goal of this study was to establish whether TLR5 signaling is the key molecular pathway mediating lung inflammation in CF, and to determine whether strategies to inhibit TLR5 can reduce the damaging inflammatory re...

TIFA as a crucial mediator for NLRP3 inflammasome.

Toll-like receptor-mediated NF-κB activation is a major innate immune reaction of vascular endothelial cells (ECs) in response to prooxidative and proinflammatory stimuli. We identified that TNF-α receptor-associated factor-interacting protein with a forkhead-associated domain (TIFA) is a regulator of priming (signal 1) and activating (signal 2) signals of nucleotide oligomerization domain-like...

The Role of the NLRP3 Inflammasome in Fibrosis

Fibrosis leads to the deposition of collagens in organs and tissues. The resulting pathology induces a loss of function in the organ it is manifested in and this loss of function modulates the morbidity and mortality in that individual. Indeed, approximately 45% of all deaths in the Western world can be attributed to fibrosis and there are no FDA approved drugs for the treatment of fibrosis. Th...
